    1. General introduction to lipid excipients and applications: 

    Lipid excipients are a group of excipients derived from fats, including oils, waxes, phospholipids, and other lipid derivatives. They are widely used in pharmaceutical formulation and manufacturing with the following applications:

     

    Solid dosage forms

    • Lubricants/Glidants: Reduce friction between the tablet and the punches and dies during the tableting process, creating a glossy and aesthetically pleasing appearance for the finished tablets.
    • Sustained-release matrix excipients: The lipid matrix helps control the release of the active pharmaceutical ingredient (API) through a diffusion mechanism (non-swelling or eroding).
    • Taste-masking excipients: Mask the taste of bitter APIs by forming a thin lipid film around them, isolating the API from the taste receptors that perceive bitterness.
    • Base system for suppositories.

    Liquid dosage forms (injectables, oral liquids)

    • Oil carriers.
    • Formulation of emulsifying and self-emulsifying drug delivery systems (SMEDDs/SNEDDs).
    • Formulation of nanostructured targeted delivery systems such as nanoemulsions, SLNs (Solid Lipid Nanoparticles), liposomes, etc.
    • Improving solubility and bioavailability for poorly soluble/poorly permeable APIs.
    • Thickening, suspending, and stabilizing APIs.

    Semi-solid dosage forms (soft gelatin capsule fills, topical formulations)

    • Carriers for the oil phase.
    • Improving solubility and bioavailability for poorly soluble/poorly permeable APIs.
    • Enhancing skin permeability for topical drugs.
    • Thickening, suspending, and stabilizing APIs.

    Sustained-release agentsGlyceryl dibehenate, cetyl alcohol, stearyl alcohol
    • Creates a matrix frame that controls API release via a diffusion mechanism; non-swelling and non-eroding.
    • Minimizes dose dumping (rapid burst release).
    Taste-masking excipientsGlyceryl dibehenate, Glyceryl monostearate
    • Effective taste masking; can be used for various APIs without requiring specific binding.
    Oil carriersMCT oil, soybean oil, castor oil
    • Increases solubility and bioavailability.
    • Supports nutritional supplementation for the product (e.g., MCT oil)
    Thickening and suspending agentsHydrogenated castor oil, cetyl alcohol, stearyl alcohol, cetostearyl alcohol, glyceryl monostearate, various waxes
    •  Increases viscosity, helping to adjust the physical consistency of liquid and semi-solid dosage forms, thereby suspending and stabilizing the API more effectively.

    • Improves solubility and bioavailability for poorly soluble APIs.
    • Used in formulating targeted drug delivery systems and reducing side effects.
