Explore Avocadoflex®, a standardized avocado-soy unsaponifiable ingredient, its phytosterol profile, research background and applications in joint health supplements.
Avocadoflex® is an Avocado-Soybean Unsaponifiables (ASU) ingredient derived from the unsaponifiable fractions of avocado fruit oil (Persea americana) and soybean oil (Glycine max). According to published product information, Avocadoflex® has a standardized phytosterol composition with an avocado-to-soybean phytosterol ratio of 1:2 and contains at least 30% total phytosterols. Its main identified phytosterols include β-sitosterol, campesterol, and stigmasterol.
ASU is a group of ingredients that has been investigated for joint health, particularly in the context of osteoarthritis. Research has focused on its potential effects on inflammatory responses, cartilage matrix metabolism, and symptoms associated with joint function [3, 4].
With its phytosterol-rich profile and plant-derived lipid composition, Avocadoflex® may be considered for nutraceutical formulations targeting joint function and mobility support. However, evidence concerning ASU as a general ingredient group should be distinguished from research conducted specifically on Avocadoflex®.
2. Mechanism of Action and Functional Properties of Avocadoflex®
Effects on Inflammation-Related Factors
In osteoarthritis, pro-inflammatory cytokines such as interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-α) are associated with an imbalance in cartilage tissue metabolism. These mediators may stimulate chondrocytes to produce inflammatory mediators and enzymes involved in extracellular matrix degradation.
Research on ASU suggests that this ingredient group may influence certain signaling pathways and mediators involved in inflammatory responses. Proposed mechanisms include effects on pro-inflammatory cytokines and prostaglandin E2 (PGE2). These findings provide a scientific basis for investigating the potential of ASU in supporting the management of osteoarthritis-related symptoms [5].
Notably, a 2026 study using a rat model of osteoarthritis directly evaluated the commercial ASU preparation Avocadoflex® (designated as CHL in the study). The researchers investigated markers associated with inflammation, matrix degradation, and cartilage structure. However, this was a preclinical animal study and cannot establish therapeutic or preventive efficacy in humans.
Effects on Cartilage Matrix Metabolism
Articular cartilage contains an extracellular matrix (ECM), including type II collagen and aggrecan. These components are important for maintaining cartilage structure and mechanical properties.
During osteoarthritis, matrix metalloproteinases (MMPs), particularly MMP-3 and MMP-13, contribute to the degradation of cartilage matrix components. Some ASU studies have reported potential effects on the activity of these enzymes and on factors involved in matrix synthesis and regulation.
Preclinical research on Avocadoflex® has also investigated markers associated with catabolic and anabolic processes in cartilage tissue. Nevertheless, these findings should be interpreted cautiously. Changes in biological markers in experimental models do not establish that the ingredient can regenerate or repair human articular cartilage.
3. Applications of Avocadoflex® in Formulation Development
Avocadoflex® may be considered for nutraceutical formulations intended to support joint function. Verified commercial products demonstrate its use in both capsule and sachet powder formats. Dosage-form selection should be based on the ingredient's technical characteristics, product objectives, and available supplier data.
A. Hard Capsules
Hard capsules are a suitable dosage-form option for formulations containing Avocadoflex® as a powder or powder blend. Commercial products listing Avocadoflex® in their capsule ingredients demonstrate that this format has already been used in the market [7, 8].
In these formulations, Avocadoflex® serves as the ASU-based active ingredient. The amount used should be determined according to the ingredient specification, formulation objectives, and relevant supporting data. The amount used in one commercial product should not automatically be treated as a universal recommended dose for other formulations.
From a formulation development perspective, powder properties such as flowability, dispersibility, and fill-weight uniformity should be evaluated. If technological improvement is required, excipients such as microcrystalline cellulose or colloidal silicon dioxide may be considered during formulation trials. Their suitability and use levels, however, must be established through actual formulation studies rather than inferred solely from the ASU composition.
The stability of the ingredient under the intended storage conditions, compatibility with other formulation components, and packaging requirements should also be assessed. These factors should be evaluated using stability data and the supplier's technical documentation.
B. Sachet Powder Formulations
Sachet powder is another application format documented in commercial products. For example, Swissmed® Opti-Flèx declares 50 mg of Avocadoflex® Avocado-Soybean Phytosterols per sachet, combined with Bonepep®, AquaLOX® Boswellia Extract, and multivitamins [9].
In this format, Avocadoflex® is used as a joint-health ingredient within a multi-ingredient formulation. During the development of sachet powders, key considerations include dispersibility, blend uniformity between sachets, and compatibility with other ingredients.
For multi-ingredient formulations, sensory properties, stability, and content uniformity should be evaluated in the finished formulation. Compatibility should not be assumed simply because the same ingredients appear together in another commercial product.
Combination with Other Joint Health Ingredients
Avocadoflex® may be considered for multi-ingredient formulations targeting joint health. Harker Herbals Joint Protect & Repair combines Avocadoflex® with Boswellia, Fermeric®, and Gotu Kola, while Swissmed® Opti-Flèx combines it with Bonepep®, AquaLOX® Boswellia Extract, and multivitamins.
These examples demonstrate that Avocadoflex® can be incorporated into combination formulations rather than being used only as a standalone ingredient. However, the presence of multiple ingredients in a commercial product does not, by itself, demonstrate synergistic effects.
For R&D teams, the functional objective of each ingredient, formulation compatibility, stability, and blend uniformity should be assessed. Claims of synergistic activity or improved efficacy require appropriate evidence from the specific formulation being developed.
The following products were selected because their published ingredient information explicitly identifies Avocadoflex®. Dosage forms and ingredient amounts are reported according to the cited manufacturer or product pages.

Simethicone is a specific mechanical defoaming agent, distinguished by its absolute physiological inertness. However, the full potential of this active ingredient is optimally harnessed when utilized in a chewable tablet dosage form. By leveraging the mechanical crushing action in the oral cavity as a "preliminary release" step, chewable tablets allow Simethicone to bypass the waiting time required for disintegration, enabling it to instantly disperse into microdroplets within the gastric fluid and initiate a rapid defoaming effect.
